What Does the Research Actually Say About Nicotine-Free Pouch Ingredients?
A review of the published evidence behind the three most common approaches to replacing nicotine in an oral pouch: caffeine, caffeine's primary metabolite, and functional mushrooms.
The nicotine-free pouch category has grown quickly, and so has the range of ingredients used to replace nicotine's stimulation. Most of that growth has outpaced the research base behind any single formula.
This isn't a review of marketing claims. It's a review of what the underlying human research shows for each ingredient category currently used across the market, evaluated on its own terms.
Three stimulant categories, three different evidence bases. Each is reviewed independently below.
Category 1: Caffeine
Caffeine is the most-studied stimulant in this category by a wide margin, with decades of human research behind its acute effects on alertness and cognitive performance. It's also the most predictable in its downsides: dose-dependent jitteriness, tolerance with regular use, and a well-documented crash as blood levels decline.
Caffeine's research base is not in question. What's relevant here is simpler: a caffeine pouch is a caffeine delivery product. The stimulant profile is nicotine's, replaced with a different, well-characterized stimulant.
Caffeine's acute effects on alertness and cognitive performance are well established in the research literature. Its side effect profile, including jitteriness, tolerance, and rebound fatigue, is equally well documented.
Category 2: Paraxanthine
Paraxanthine is caffeine's primary liver metabolite, produced when the CYP1A2 enzyme breaks caffeine down. Roughly 84 percent of ingested caffeine converts to paraxanthine in the body.1 Because it occurs naturally as a byproduct of caffeine metabolism, some formulations use it directly, positioning it as a "caffeine-free" alternative on the theory that it produces similar alertness with a shorter half-life.
The research on paraxanthine specifically, separate from caffeine, is newer and considerably smaller than caffeine's own literature. Early data suggests a faster clearance time compared with caffeine, but the ingredient is still, mechanistically, part of the same stimulant pathway.1

Lion's Mane is included in functional mushroom formulas for focus and cognitive support. In a randomized, double-blind, placebo-controlled trial of 41 healthy adults, a single dose produced significantly faster performance on a standard cognitive test (the Stroop task) compared with placebo, with a trend toward reduced subjective stress after 28 days of continued use.2 The trial's authors note the sample size was small and describe the findings as a promising early signal rather than a settled conclusion.

A randomized, placebo-controlled trial found that three weeks of a Cordyceps-containing mushroom blend significantly improved VO2 max, a standard measure of the body's ability to use oxygen during exercise, compared with placebo.3 A separate randomized crossover trial found that Cordyceps militaris improved reaction time following exhaustive exercise compared with placebo.4 Both trials involved relatively small samples and measured physical or cognitive performance markers rather than subjective energy directly, so the research speaks most confidently to exercise- and blood-flow related outcomes.

Reishi is included for stress and wellness support. In a randomized, double-blind, placebo-controlled trial of 132 adults with chronic fatigue, an 8-week course of Reishi extract improved fatigue scores and overall sense of well-being compared with placebo.5 The study population had diagnosed chronic fatigue rather than being a general healthy population, which is worth noting when applying the finding more broadly.
What the Evidence Supports, And What It Doesn't
Does this mean functional mushrooms are proven to outperform caffeine or paraxanthine?
A: No. The three ingredient categories have not been tested head-to-head in a single controlled trial. What the research supports is narrower: each mushroom ingredient has its own independent evidence for the specific effect it's included for, through a mechanism that isn't stimulant-based.
Are these findings definitive?
A: No single study here is large enough to be called definitive. Sample sizes range from a few dozen to just over 100 participants, several trials measured acute or short-term effects rather than long-term use, and some findings (like Lion's Mane's stress trend) fell just short of standard statistical thresholds. They represent a genuine, growing evidence base, not a closed case.

Where This Leaves the Comparison
Caffeine and paraxanthine both work by acting on the same general stimulant pathway nicotine also acts on, just through different specific compounds. Functional mushrooms work through separate, non-stimulant mechanisms, each with its own independent research.
Neither approach is unproven. They're evaluated on different kinds of evidence, and it's worth knowing which kind you're actually getting when you read a pouch label.
